SILAC and mTRAQ comparison for phosphoproteomics on Q-exactive


Most people are familiar with iTRAQ labeling but only few know about mTRAQ. There are studies that compare between SILAC and iTRAQ  as well as iTRAQ and mTRAQ quantitation methods. Now, this recent article by in JPR talks about SILAC and mTRAQ comparison (article). As both of these methods quantitate at MS level, it is interesting to see their comparison unlike iTRAQ and SILAC.

Overall, the mTRAQ method identified relatively more proteins than SILAC but as far as variation in quantitation is concerned, SILAC gave better results than mTRAQ. The take home message here is that mTRAQ can be used as a substitute to metabolic labeling methods such as SILAC. Apart from this comparison, they also studied anti-oncogenic mechanism downstream of ALK kinase in lung cancer cells.
