I thought I posted this a while back when I first learned about it, but I guess I didn't, cause I couldn't find it when I went looking for it!
I call this the EasyNano Super Wash. To give proper credit, I first learned this last summer from Dr. Craig Dufresne, though it has probably been adapted a little since I generally can not read my own hand writing.
Sometimes we get carry over. It is a weakness of nanoLC. We're moving insanely low volumes around and, let's face it, sometimes you are injecting sticky junk into your instrument. This means that sometimes the standard wash doesn't cut it.
What you need is a SuperWash! Ignore the volumes above.
Do this!
In wash bottle 1 put buffer A
In wash bottle 2 put 98% buffer B (or even better 98% methanol! wooo!)
In wash bottle 3 put isopropanol, at least 80%
Switch your wash from Standard to Custom (the only reason to copy what is in the picture above. Don't put in those numbers. They won't work unless you are using an imaginary 50 uL loop. You'll just get an error message, and then you'll forget to take a screenshot of a method that will work!)
Do a wash with bottle 1, full loop
Do a wash with bottle 2, full loop
Do a wash with bottle 3, full loop
Do 2 washes with bottle 2
Do 3 washes with bottle 1, your buffer A.
Boom! You've washed the crap out of your needle, your loop and the isopropanol has gotten rid of any residual air introduced by that guy who never makes sure there aren't air bubbles at the bottom of his autosampler vials.
